Principle of the Assay: The Picokine Human ANGPTL6 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human ANGPTL6 with a 96-well strip plate that is pre-coated with antibody specific for ANGPTL6. The detection antibody is a biotinylated antibody specific for ANGPTL6. The capture antibody is A monoclonal antibody from mouse, the detection antibody is a biotinylated detection polyclonal antibody from goat. The kit contains recombinant Human ANGPTL6 with immunogen: Expression system for standard: CHO, Immunogen sequence: Ala20-Leu470. The kit is analytically validated with ready to use reagents. To measure Human ANGPTL6, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human ANGPTL6 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human ANGPTL6 in the sample!!Background/Introduction: Angiopoietin-like 6 (ANGPTL6), also known as angiopoietin-related growth factor (AGF), is a secreted 50 kDa protein that contains a coiled-coil domain and a fibrinogen-like domain. It is mapped to 19p13.2. ANGPTL6 is highly expressed in hepatocytes and is detectable in serum. Within the liver, secreted ANGPTL6 inhibits gluconeogenesis and promotes insulin sensitivity and energy expenditure. Circulating ANGPTL6 promotes the chemotaxis of vascular endothelial cells resulting in increased vascular permeability and angiogenesis. An integrin?-binding RGD motif located within the fibrinogen-?like domain of ANGPTL6, enables it to promote skin wound healing by mediating the adhesion and migration of keratinocytes, fibroblasts, and endothelial cells. Elevated levels of ANGPTL6 in serum are associated with metabolic syndrome, diabetes, and preeclampsia. Serum ANGPTL6 levels are also elevated during obesity. ANGPTL6 is reported to promote the survival and expansion of hematopoietic stem cells. ANGPTL6 is produced by several hematopoietic cell types including megakaryocytes, platelets, mast cells, and uterine NK cells. Mature human ANGPTL6 shares 75% sequence identity with both mouse and rat ANGPTL6.
