Principle of the Assay: The Picokine Rat Cdh3 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Rat Cdh3 with a 96-well strip plate that is pre-coated with antibody specific for Cdh3. The detection antibody is a biotinylated antibody specific for Cdh3. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Rat Cdh3 with immunogen: Expression system for standard: NSO, Immunogen sequence: E100-G647. The kit is analytically validated with ready to use reagents. To measure Rat Cdh3, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Rat Cdh3 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Rat Cdh3 in the sample!!Background/Introduction: Cadherins are calcium-dependent cell-cell adhesion molecules that mediate cell-cell binding in a homophilic manner. They play an important role in the growth and development of cells via the mechanisms of control of tissue architecture and the maintenance of tissue integrity. Cadherin expression is regulated spatially as well as temporally. Cadherins are thought to play an important role in development and maintenance of tissues through selective cell-cell adhesion activity and may be involved also in the invasion and metastasis of malignant tumors. Cadherin regulates dendritic spine morphogenesis. A cadherin gene cluster is mapped to a region of chromosome 5 subject to frequent allelic loss in carcinoma. The standard product used in this kit is recombinant P-Cadherin with the molecular mass of 120-130Kda after glycosylation.
