Principle of the Assay: The Picokine™ Mouse Icam1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Mouse Icam1 with a 96-well strip plate that is pre-coated with antibody specific for Icam1. The detection antibody is a biotinylated antibody specific for Icam1. The capture antibody is monoclonal antibody from rat and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Mouse Icam1 with immunogen: Expression system for standard: NS0; Immunogen sequence: Q28-N485. The kit is analytically validated with ready-to-use reagents. To measure Mouse Icam1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Mouse Icam1 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Icam1 in the sample.
Background/Introduction: Intercellular adhesion molecule-1 (ICAM-1) is an integral membrane protein, a member of the immunoglobulin superfamily, and a ligand for lymphocyte function-associated (LFA) antigens, a beta 2 leukocyte integrin. The normal function of human intercellular adhesion molecule-1 (ICAM-1) is to provide adhesion between endothelial cells and leukocytes after injury or stress. ICAM-1 binds to leukocyte function-associated antigen (LFA-1) or macrophage-1 antigen (Mac-1). ICAM-1 is found on leukocytes, fibroblasts, epithelial cells and endothelial cells and its expression is regulated by inflammatory cytokines. ICAM-1 has a tissue distribution similar to that of the major histocompatibility complex class II antigens and is likely to play a role in inflammatory responses. The standard product used in this kit is produced by recombinant gene expression, which is a dimeric protein linked with disulfide bonds. The single chain is Gln28-Asn 485. It has 457 amino acids with the molecular mass of 76.8kDa. As a result of glycosylation, the molecular mass of 110-130KDa is revealed by SDS-PAGE.
