Background: Thromboxane B2 is a prostaglandin derivative that is released during anaphylaxis. Thromboxane B2 induces arterial contraction and platelet aggregation. Since platelets participate in atherogenesis and contribute to acute, ischemic complications, elevated TxB2 levels may reflect ongoing cardiovascular, peripheral vascular, and cerebrovascular disease processes. TxB2 is the stable, inactive product of prostaglandin metabolism of thromboxane A2, which is renally cleared and, therefore, can be measured in the urine.
Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been pre-coated with TXB2. During the reaction, TXB2 in the sample or standard competes with a fixed amount of TXB2 on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to TXB2. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzymesubstrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of TXB2 in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
