Background: syndecan-1 is a member of the transmembrane heparan sulfate proteoglycan family. syndecan-1 is expressed in most epithelial cells and its expression is highly regulated and specific to cell type and stage of development. Through its extracellular region, the side chain can covalently bind a series of ligands, regulate the interaction between cells and the microenvironment as asce11-surface co-receptors, and play an important role in the support, adhesion, movement, proliferation and differentiation of normal cells. The expression of syndecan-1 decreased in various tumor cells, which is closely related to the growth, differentiation and metastasis of tumor cells, and can be used as an indicator to judge the prognosis of various tumors.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti SDC1 antibody was pre-coated onto the 96-well plate. The biotin conjugated anti SDC1 antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with SDC1 conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of SDC1 in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
