Background: CGRP is the strongest vasodilator substance known, which has the effect of decreasing blood pressure, decreasing peripheral resistance, renal artery dilation, and significantly increasing renal blood flow. CGRP also has A strong diastolic effect on coronary arteries, and is also effective on atherosclerotic coronary arteries. This diastolic effect is independent of the presence of vascular endothelium, and is not affected by A, B and 5-hydroxytryptamine receptor blockers. CGRP can also reduce coronary perfusion pressure, significantly increase coronary blood flow, accelerate heart rate, and enhance myocardial contractile force. CGRP has obvious positive muscle strength and positive chronotropic effect, which is stronger than norepinephrine, but can be cancelled by B receptor blockers.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti CGRP antibody was pre-coated onto the 96-well plate. The biotin conjugated anti CGRP antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with CGRP conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of CGRP in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
