Background: IL-1b(Interleukin-1beta) is a cytokine produced primarily by macrophages, monocytes, and other immune cells. It is a key mediator in inflammatory responses, promoting the occurrence and development of inflammation and participating in the immune and inflammatory response of the body. By binding to its receptor, IL-1b can activate various signaling pathways, inducing inflammatory reactions and activation and proliferation of immune cells. Additionally, IL-1b can induce the production of other inflammatory and cytokine factors by various cells, such as TNF-alpha and IL-6, further exacerbating the inflammatory response. Abnormal expression and signaling pathways of IL-1b are associated with many autoimmune diseases, infectious diseases, and hypersensitivity diseases.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti IL-1beta antibody was precoated onto the 96-well plate. The biotin conjugated anti IL-1beta antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with IL-1beta conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of IL-1beta in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
