Background: beta-defensin-2 (HbetaD-2) is a small cysteine-rich cationic skin-antimicrobial peptide (SAP) produced by a number of epithelial cells.beta-defensin-2 has antimicrobial activity against gram-negative bacteria and Candida, but not gram-positive Staphylococcus aureus. beta-defensin-2 can be used for the study of colitis.
Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been pre-coated with Defb2. During the reaction, Defb2 in the sample or standard competes with a fixed amount of Defb2 on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to Defb2. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of Defb2 in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
