Background: KIM-1 is also a biomarker of AKI in humans. There was extensive expression of KIM-1 in proximal tubule cells in kidney biopsies from all six patients with biopsy-confirmed ATN.KIM-1 is also a sensitive biomarker of tubular injury in other renal diseases besides AKI. KIM-1 was primarily expressed at the luminal side of dedifferentiated proximal tubules, in areas with fibrosis and in areas of inflammation in macrophages. KIM-1 is upregulated in renal disease and is associated with renal fibrosis and inflammation and that urinary KIM-1 can be used as a noninvasive biomarker of tubular injury in multiple renal diseases.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti KIM-1 antibody was precoated onto the 96-well plate. The biotin conjugated anti KIM-1 antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with KIM-1 conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of KIM-1 in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
