Background: Heat Shock Protein 72 (HSP72), also known as Heat Shock Protein 1A (HSPA1A), is a member of the HSP70 family of molecular chaperones. HSP72 promotes the proper folding of nascent polypeptides and assists in the refolding of denatured proteins. However, if either of these processes proceeds too slowly or fails, HSP72 can interact with the HSP40 co-chaperone protein and the CHIP/STUB1 Ubiquitin ligase (E3) to promote ubiquitination and degradation of the nascent polypeptide or denatured protein. Pathologically, HSP72 has been implicated in the promotion of multiple cancer types. Conversely, it is thought to protect against several neurodegenerative diseases that are caused by the accumulation of misfolded proteins.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti HSP72 antibody was pre-coated onto the 96-well plate. The biotin conjugated anti HSP72 antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with HSP72 conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of HSP72 in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
