The thiol groups in living organisms mainly include glutathione thiol and protein thiol. The former not only repairs proteinsdamaged by oxidation, but also participates in the clearance of reactive oxygen species, while the latter plays an important role inmaintaining protein conformation. By measuring the total sulfhydryl group content and glutathione content, protein thiol contentcan be indirectly determined. The thiol group reacts with 5, 5'- dithio bis nitrobenzoic acid (DTNB) to form a yellow compound witha maximum absorption peak at 412 nm.
