Recombinant human AAK1 (1-465) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. The gene accession number for AAK1 is BC002695.
Adaptor-associated kinase 1 (AAK1) is a serine/threonine protein kinase of the NAK kinase. Along with GAK, AAK1 regulates clathrin-mediated endocytosis by phosphorylating the u subunits of both AP-1 and AP-2 trafficking adaptors. The phosphorylation increases the affinity of AP2 for membranes and induces a conformational change enabling cargo recruitment by this adaptor. AAK1 was also shown to play a positive role in regulating the Notch pathway.
