- Cortactin is an ~80–85 kDa actin-binding protein that orchestrates cytoskeletal remodeling and signal integration. It contains an N-terminal repeat domain that associates with filamentous actin, followed by a helical segment, a proline-rich region with multiple signaling motifs, and a C-terminal SH3 domain. Through interactions with the Arp2/3 complex and other regulators, cortactin promotes actin branching, lamellipodia formation, and endocytic trafficking. Tyrosine phosphorylation by Src family kinases inhibits its actin crosslinking activity, linking receptor-mediated signals to cytoskeletal rearrangements. Aberrant cortactin activity has been implicated in tumor cell invasion, metastasis, and vascular permeability disorders, highlighting its importance in both normal cellular dynamics and disease progression.
