SON is a ubiquitously expressed DNA- and RNA-binding protein primarily localized to nuclear speckles. SON involves in multiple cellular processes including transcription, pre-messenger RNA (mRNA) splicing, and cell cycle regulation. SON is a key component of the spliceosomal complex and a critical mediator of constitutive and alternative splicing. During constitutive RNA splicing, SON ensures efficient intron removal from the transcripts containing suboptimal splice sites. Importantly, SON-mediated splicing is required for proper processing of selective transcripts related to cell cycle, microtubules, centrosome maintenance, and genome stability. Moreover, SON regulates alternative splicing of RNAs from the genes involved in apoptosis and epigenetic modification. In addition to the role in RNA splicing, SON has the ability to suppress transcriptional activation at certain promoter/enhancer DNA sequences. Deleterious variants in SON are associated with a severe multisystem disorder characterized by developmental delay, persistent feeding difficulties, and congenital malformations, including brain anomalies. SON plays important roles in nuclear organization and pre-mRNA splicing as well as maintaining cellular health. SON has a key role in stem cells as well as during the onset of various diseases such as cancer, influenza, and hepatitis.
