Function: Orphan receptor. Signals via G(s)-alpha family of G-proteins (PubMed:22025619, PubMed:22575658). Has protumorigenic function especially in glioblastoma (PubMed:27775701).
Similarity: A short peptide sequence (termed the Stachel sequence) in the C-terminal part of the extracellular domain (ECD) functions as a tethered agonist. Upon structural changes within the ECD, e.g. due to extracellular ligand binding or mechanical movements, this intramolecular agonist is exposed to the 7 transmembrane region, triggering G-protein activation.The N-terminal domain and autocatalytic activity of ADGRD1 at the GPCR proteolysis site (GPS) are not required for G-protein coupling activity. Belongs to the G-protein coupled receptor 2 family. Adhesion G-protein coupled receptor (ADGR) subfamily.
