IL1alpha is a proinflammatory cytokine produced by activated monocytes, lymphocytes, and epithelial cells (1). IL1alpha is synthesized as an active precursor protein that appears to be cleaved by cytosolic proteases into its mature form (1,2). Often, precursor and mature forms of IL1alpha are primarily retained intracellularly rather than constitutively secreted. (1,2). Signaling by IL1alpha involves IL1alpha binding to an IL1 accessory protein (IL1AcP) and then the complex binds to IL1RI (1,2). Signaling occurs through activation of MAP kinase and NFkappaB pathways (1,2). IL1alpha also binds to an ILRII that lacks an intracellular signaling domain and thereby serves as a high affinity decoy receptor. IL1alpha activity is inhibited through IL1R antagonist (IL1Ra) that binds IL1R1 but does not signal. IL1alpha has been shown to be a key mediator of virusinduced inflammatory responses in mice (3).
