The pooled antisera were raised in rabbits which were immunized with synthetic human PTH (39-84) covalently attached onto bovine thyroglobulin as a carrier protein, and the rabbit antibody has been characterized by RIA, immunocytochemical, western immunoblot and ELISA techniques. The antibody has been found to be highly specific for this peptide sequence and was shown to immunoprecipitate human PTH(39-84) by RIA techniques.
The IgG fraction was purified from the pooled antisera and was shown to have a purity of greater than 95%. The purified IgG was lyophilized from a solution containing a volatile buffer. Dilute the lyophilized IgG with 0.1 ml of buffer (such as phosphate buffered saline) for the equivalent IgG concentration of whole antiserum, or with additional buffer (such as 1 mg/ml BSA in PBS) for more dilute antibody. Each vial contains 1.8 mg of purified rabbit IgG.
Once re-dissolved, avoid repeated freeze/thaw cycles since the purified antibody can be denatured. We recommend aliquoting the re-dissolved IgG into smaller quantities prior to freezing. Store the aliquots frozen at -20 degree C or lower. Once an aliquot is thawed, store it at 4 degree C. We recommend that the end user perform an immuno-titration experiment (starting at a dilution of 1:500) on their sample using their specific techniques since different laboratories use various techniques.
