3D 1.1 reacts with the human Type I hPTH/PTHrP and it does not react with the Type II receptor. The antibody has been used in immunochemical studies of human ovarian tissues to demonstrate a novel role for PTH and/or PTHrP in ovarian function.
3D 1.1 recognises proteins with apparent molecular weights of 80,000 and 160,000 daltons in western blots of celllysates. Competitive binding assays show that the epitope recognized by 3D1.1 is localized to the C-terminal region of the immunogenic peptide. Furthermore, a corresponding peptide from the related PTH receptor (hPTH2) was shown to have minimal (0.1 %) cross reactivity. Finally, F ACS analysis has also been used to show that the antibody recognizes C-21 cells, but not parental BEK cells that express only the hPTH2 receptor.
Immunohistochemical staining of fixed, paraffin-embedded tissue shows specific hPTHlPTHrP receptor expression in chondrocytes, renal tubular and glomerular epithelium, afferent and efferent renal arterioles (but not glomerulr endothelium), and vascular smooth muscle and endothelium. This finding confirms that 3 D 1.1 positively stains classical PTH target tissues.
