IRAK (IL-1 associated serine/threonine kinase) is critical regulator of interleukin-1 (IL-1) induced activation of the NFkappaA pathway. IL-1 stimulation leads to recruitment of interleukin-1 receptor-associated kinase (IRAK) to the IL-1 receptor, where IRAK is phosphorylated, ubiquitinated, and eventually degraded. There are four members (IRAK1, IRAK2, IRAK3-IRAKM and IRAK4). A novel member in the IRAK/Pelle was recently identified and designated IRAK-M. IRAKs associate with IL-1/Toll receptors after IL-1 or LPS stimulation and the dominant negative mutants or IRAKs inhibit IL-1 or LPS induced NF-kB activation. Members in IRAK/Pelle family play a central role in IL-1R/TLR mediated inflammatory responses to cytokine IL-1 and LPS.
