细胞培养级猪I型胶原

      胶原蛋白是脊椎动物体内丰富的蛋白质,存在于皮肤、软骨、骨、椎间盘、血管、肌腱、韧带和角膜中,是细胞外基质(ECM)的主要成分。I型胶原由两条相同的α1链和一条不同的α2链(1)组成,形成称为原胶原的三螺旋结构。这种三螺旋构象增加了组织的结构强度和抗酶降解能力,在形成细胞外基质过程中起着关键作用。
      胶原蛋白有助于促进组织再生和/或靶向药物释放,因为它具有低抗原性,低毒、高水溶性、高生物降解性。I型胶原结合整合素,促进细胞迁移、附着以及增殖和分化。尽管通过胃蛋白酶消化从原胶原中去除N-和C-末端原胶原被广泛用于工业用途,但原胶原作为含有交联末端肽的天然胶原支架可能是有益的。
      Chondrex公司提供酸性I型原胶原溶液,可用于传统的二维(2D)系统以及三维(3D)凝胶中的支架,用于模拟成纤维细胞和软骨细胞中的细胞生长。

操作流程 包被(注:具体的包被的条件需要根据培养系统优化
1.用0.02mol/L的盐酸稀释4mg/ml的胶原到50-100ug/ml。
2.温和混匀稀释后的溶液。
3.取适量的稀释后的胶原加入孔内或板内。
注:保证整个表面都是包被的。
4. 37℃或室温孵育1-2小时
5. 弃掉所有溶液。
6.用培养基或者PBS漂洗包被表面。
7.包被好的孔或板2-8℃保存或者空气中晾干(如果保持无菌)
三维凝胶制备流程:
  1. 用PBS等体积稀释胶原溶液,终浓度2mg/ml。
  2. 加入适量的稀释后的胶原到孔内或板内。
  3. 37℃孵育30~60分钟。
  4. 凝胶可以2-8℃保存或者空气中晾干(如果保持无菌)。
