Principle of the Assay: The Picokine Mouse Serpine2 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Mouse Serpine2 with a 96-well strip plate that is pre-coated with antibody specific for Serpine2. The detection antibody is a biotinylated antibody specific for Serpine2. The capture antibody is A monoclonal antibody from rat, the detection antibody is a biotinylated detection polyclonal antibody from goat. The kit contains recombinant Mouse Serpine2 with immunogen: Expression system for standard: NSO, Immunogen sequence: S20-P397. The kit is analytically validated with ready to use reagents. To measure Mouse Serpine2, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Mouse Serpine2 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Serpine2 in the sample!!Background/Introduction: Glia-derived nexin is a protein that in humans is encoded by the SERPINE2 gene. This gene is mapped to 1 C4; 1 40.97 cM. SERPINE2 produced in Sf9 Baculovirus cells is a single,glycosylated polypeptide chain containing 386 aminoacids (20-397a.a.) andhaving a molecular mass of 42.9kDa (Molecular size on SDS-PAGE will appear atapproximately 40-57kDa).SERPINE2 is expressed with a 8 aminoacid His tag atC-Terminus and purified by proprietary chromatographic techniques. This gene displays neurite promoting activity and serine protease inhibitory activity.
