Background: Melatonin is a hormone synthesized by the pineal gland, whose secretion is regulated by light. In dark environments, the release of melatonin from the pineal gland increases, while in light, it decreases. Melatonin plays an important role in regulating the biological clock, sleep-wake rhythm, immune function, and antioxidant defense in mammals. Due to its regulatory effects, Melatonin has been widely used in the treatment of sleep disorders such as insomnia and jet lag. Additionally, Melatonin has potential medical applications in anti-aging, anti-cancer, and neuroprotection.
Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been precoated with MT. During the reaction, MT in the sample or standard competes with a fixed amount of MT on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to MT. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of MT in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
