Short Description: Recombinant JARID1C/KDM5C (accession number NP_004178.2) was expressed in Sf9 and contains an N-terminal DYKDDDDK tag with a molecular weight of 180 kDa. Lysine (K)-specific demethylase 5C (KDM5C), also known as Jumonji, AT rich interactive domain 1C (JARID1C), is a member of the SMCY homolog family and encodes a protein with one ARID domain, one JmjC domain, one JmjN domain and two PHD-type zinc fingers. It is a histone demethylase that specifically demethylates lysine 4 of histone H3 (H3K4), thereby playing a central role in defining the histone code. KDM5B demethylates trimethylated and dimethylated, but not monomethylated H3K4. It does not demethylate histone H3K9, H3K27, H3K36, H3K79 or H4K20. KDM5C participates in transcriptional repression of neuronal genes by recruiting histone deacetylases and CoREST pAb at neuron-restrictive silencer elements. Mutations in KDM5C have been associated with X-linked mental retardation. Recombinant JARID1C/KDM5C can be used for enzyme kinetics and inhibitor screening studies.
