This antibody recognizes human, mouse, and rat ADAM10. A band of 85 kD is detected in immunoblots and may represent precursor. A 60 kD band is detected in some cell lines, including Jurkat and appears to be the processed mature protein.

Tumor necrosis factor alpha (TNF alpha ) is a proinflammatory cytokine and contributes to a variety of inflammatory reactions and to programmed cell death. Notch receptor and its ligand participate in cell fate decisions during vertebrate development and are associated with several human disorders, including T-cell lymphoma. TNF alpha , Notch and its ligand delta are all membrane-bound molecules which are cleaved by proteases to release mature proteins or functional receptors. ADAM10, a metalloproteasedisintegrin in the famioly of mammalian ADAM (for a disintegrin and metalloprotease) was recently found to cleave TNF alpha , Notch and its ligand delta. The genes encoding human, mouse, and bovine ADAM10 have been cloned and designated ADAM10, Kuzbanian (KUZ), and MADM, respectively. These genes are expressed in a variety of human and bovine tissues.
