CD163 is an acute phase-regulated receptorinvolved in clearance and endocytosis ofhemoglobin/haptoglobin complexes bymacrophages and may thereby protect tissuesfrom free hemoglobin-mediated oxidativedamage. The protein may play a role in theuptake and recycling of iron, via endocytosis ofhemoglobin/haptoglobin and subsequentbreakdown of heme. It bindshemoglobin/haptoglobin complexes in acalcium-dependent and pH-dependent manner.And it exhibits a higher affinity for complexes ofhemoglobin and multimeric haptoglobin of HP*1Fphenotype than for complexes of hemoglobinand dimeric haptoglobin of HP*1S phenotype. Italso induces a cascade of intracellular signalsthat involves tyrosine kinase-dependent calciummobilization, inositol triphosphate production andsecretion of IL6 and CSF1.
